Immunological determination of triazine pesticides bound to soil humic acids (bound residues)

An immunological method for the determination of triazine herbicides covalently bound to soil humic acids has been developed. A sandwich-immunoassay has been performed, based on both polyclonal humic acid-antibodies and monoclonal triazineantibodies. A peroxidase-labelled third antibody has been used for the photometric detection. A triazine-humic acid conjugate served as calibration standard. The coupling density for this conjugate has been determined by measuring the difference of free amino groups both with ninhydrin and with the trinitrobenzene sulfonic acid method. In addition, the coupling density has been confirmed by scintillation counting using a (14)C-atrazine derivative. Due to nonspecific interactions between antibody proteins and humic acids, different blocking steps had to be performed. Finally, the assay has been applied to a triazine contaminated soil sample. Humic acids (including bound residues) have been extracted by diluted sodium carbonate solution. Concentrations of bound atrazine residues have been found in the range of 2 mg/kg soil on fields where triazine herbicides has been applied over a period of 21 years. These results are comparable to both the applied amount and the nonextractable fraction.

We consider the quantum mechanical problem of a particle being scattered inelastically by a chain ofN infinitely heavy, equidistantly spaced two-level atoms. In a previous paper (Süßmann, G., Szilas, P.: Z. Phys. B — Condensed Matter42, 253 (1981)) the time dependent problem of a Gaussian wave packet impinging on the target atoms has been studied and an explicit asymptotic expression for the reduced density matrix ρ _R of the particle has been given. We now introduce the coarse grained density matrix \(\bar \rho _R\) . The incoherenceI?1-Tr( \(\bar \rho _R^2\) ), i.e. the deviation of the state of the particle from a pure state, being small on certain conditions, we find a single particle wave function ψ with \(\psi (x',t)\psi ^* (x'',t) \approx \left\langle {x'\left| {\bar \rho _R (t)} \right|x''} \right\rangle\) and a nonlinear Hermitean HamiltonianH _ψ=p ²/2m+W _ψ such thati?ψ(x, t)=H _ψψ(x, t) describes the time evolution. Finally we also considerW _ψ within the framework of the phenomenological theory of nonlinear frictional operators.     

Nonaqueous capillary electrophoresis with indirect electrochemical detection

Nonaqueous capillary electrophoresis (NACE) which makes use of organic solvents in place of conventional aqueous electrophoresis buffers is gaining increasing importance among modern separation techniques. Recently, it has been shown that amperometric detection in conjunction with acetonitrile-based NACE offers an extended accessible potential range and an enhanced long-term stability of the amperometric responses generated at solid electrodes. The present contribution takes advantage of the latter aspect to develop reliable systems for NACE with indirect electrochemical detection (IED). In this context, several compounds such as (ferrocenylmethyl)trimethylammonium perchlorate, tris(1,10-phenanthroline)cobalt(III) perchlorate and bis(1,4,7-triazacyclononane)nickel(II) perchlorate were studied regarding their suitability to act as electroactive buffer additives for IED in NACE. The performance characteristics for the respective buffer systems were evaluated. Tetraalkylammonium perchlorates served as model compounds for the optimization of the NACE-IED system. Target analytes choline and acetylcholine could easily be separated and determined by means of NACE-IED. In the case of a buffer system containing 10(-4) M tris(1,10-phenanthroline)cobalt(III) perchlorate the limits of detection were 2.5 x 10(-7) M and 4.6 x 10(-7) M for choline and acetylcholine, respectively. With the elaborated analytical procedure choline could be determined in pharmaceutical preparations.     

Standardisierung der dünnschichtchromatographischen Trennungen von Flechtensäuren zur Chemotaxonomie von Flechten

Zusammenfassung Die dünnschichtchromatographische Trennung von Flechtenextrakten zur Unterstützung der Taxonomie wurde in Bezug auf die Trennleistung und auf die Reproduzierbarkeit optimiert. Dabei wurden besonders gute Trennleistungen erhalten mit dem Flie?mittelgemisch Benzol:Dioxa:Eisessig=90∶25∶4 bei Verwendung von Kieselgel, dessen Aktivit?t durch eine Umgebung mit relativer Feuchte von 70% festgelegt wurde, und mit Methylenchlorid als Eluiermittel bei Verwendung von mit Oxals?ure impr?gniertem Kieselgel und antiparallelem Aktivit?tsgradienten. Eine hohe Reproduzierbarkeit der Trennungen wurde erreicht durch Verwendung der Vario-KS-Kammer nach Geiss, die die Einstellung einer definierten Schichtaktivit?t bzw. eines definierten Aktivit?tsgradienten und eine reproduzierbare Vorbedampfung der Schicht mit Flie?mittel erm?glicht. Es konnte gezeigt werden, da? durch unterschiedliche chromatographische Trennungen der gleichen Extrakte und durch die Verwendung verschiedener Anf?rbereagenzien die Identifizierung und damit die Aussagekraft für taxonomische Probleme steigt.

Standardisation of the thin-layer chromatographic separation of lichen acids for the chemotaxonomy of lichens

Summary The thin-layer chromatography of lichen extracts for taxonomic studies has been optimized in relation to separation and to reproducibility. Especially good separations were found with benzene:dioxane:acetic acid (90∶25∶4) developer and silica gel thin-layer plates, whose activity was fixed by an ambient moisture content of 70%, and with methylene chloride developer and silica gel plates impregnated with oxalic acid and antiparallel activity gradient. High reproducibility of separation was obtained using the Geiss Vario-KS tank, which allows the activity of the plates or the activity gradient to be adjusted and which also allows reproducible exposure of plates to the vapour of developing solvents. By different chromatographic separations of the same extracts and by using different colour reactions the accuracy of identification for taxonomic studies has been improved.

     

Validation studies and proficiency testing

Genetically modified organisms (GMOs) entered the European food market in 1996. Current legislation demands the labeling of food products if they contain <1% GMO, as assessed for each ingredient of the product. To create confidence in the testing methods and to complement enforcement requirements, there is an urgent need for internationally validated methods, which could serve as reference methods. To date, several methods have been submitted to validation trials at an international level; approaches now exist that can be used in different circumstances and for different food matrixes. Moreover, the requirement for the formal validation of methods is clearly accepted; several national and international bodies are active in organizing studies. Further validation studies, especially on the quantitative polymerase chain reaction methods, need to be performed to cover the rising demand for new extraction methods and other background matrixes, as well as for novel GMO constructs.     

Final elucidation of the absolute configuration of the signal metabolite hormaomycin

The complete absolute configuration of hormaomycin 1 a has been established by HPLC and HPLC/MS experiments with appropriately derivatized 4-propylprolines, (2S,4S)-6 and (2R,4R)-6, as well as 4-(Z)-propenylprolines, cis-5 and trans-5, and also feeding experiments with enantiomerically pure samples of the deuterium-labeled 3-(2'-nitrocyclopropyl)alanine, (2S)-3,3-[D2]15 and (2S)-2,2'-[D2]15, and 4-(Z)-propenylproline 2',4-[D2]-(2S,4R)-5. The latter five amino acids were prepared for the first time and allowed one to unequivocally assign the hitherto unknown absolute configurations of the last four stereocenters in hormaomycin 1 a. As a bonus, some new information about the biosynthesis of this molecule has also been gathered.     

Metallorganische Verbindungen des Kupfers XIII. Kupfer(I)-chlorid-Komplexe winkelgespannter Cycloheptine Synthesen und Röntgenstrukturanalysen

The strained cycloheptynes 2a, 2b and 2c react with copper(I) chloride to form the dinuclear complexes [CuCl(cycloheptyne)]₂ 3a, 3b and 3c respectively. X-ray diffraction studies on all three compounds 3 show an increase in strength of the copper-alkyne bond with increasing deformation of the alkyne moiety from the ideal 180°. Thus going from the least strained alkyne (2c) to the most strained one (2a), the Cu-C bond lengths in the corresponding complexes decrease from 197.5 pm (3c) to 194.9 pm (3a). The C≡C-C angles increase from 145.6° (3a) over 147.4° (3b) to 151.6° (3c).

Die gespannten Cycloheptine 2a, 2b und 2c reagieren mit Kupfer(I)—chlorid unter Bildung der zweikernigen Komplexe [CuCl(cycloheptin)]₂ 3a, 3b und 3c. Röntgenstrukturanalysen von allen drei Verbindungen 3 zeigen, daβ die Stärke der Kupfer—Alkin-Bindung zunimmt je mehr das ganze Alkinsystem vom idealen 180°-Winkel abweicht. Geht man vom am wenigsten gespannten Alkin (2c) zum gespanntesten (2a), so nehmen die Cu-C-Bindungslängen in den entsprechenden Komplexen von 197.5 pm (3c) auf 194.9 pm (3a) ab. Die C≡C-C-Winkel werden von 145.6° (3a) über 147.4° (3b) nach 151.6° (3c) hin gröβer.

Summary Nitrite can be detected sensitively and specifically by spotting on Chromatograph paper impregnated with brucine, benzidine and phosphoric acid. It is possible to detect 20g nitrite in the presence of nitrate and other anions.
